Abstract

To meet these challenges we aim to: 1) Computationally optimize, synthesize and validate in vivo novel leads that allosterically modulate Hsp90 functional motions and induce cancer cell death through different mechanisms than ATP-competitive drugs. 2) Develop integrative computational methods to elucidate the key determinants of Hsp90 recognition, and translate this information into the evolution of chemical tools targeting select PPIs. 3) Map the effects of designed molecules on physiological assemblies in vitro and in vivo, and evaluate synergistic effects with known drugs in the context of drug-resistant cell lines.
